How to prepare an object for microscopy

In order to look at something throug a microscope, the object must confirm to certain criteria:

- The object must be so that the details you want to see can be illuminated.

- The object must have properties so that one can focuse sharply at it

- An object must have a size so that it fits under the microscope

- The objective must be protected from contamination form the bobject

- If the object shall be used many times, it must be prepared so that it can be stored

 

Preparing a fresh liquid object

If you want to look at something liquid or floating in its natural shape, for example a drop of water with microorganisms and algae in it, the preparation can be very simple.

You just take a drop of the liquid onto the object glass and lay a cover glass over it. Then you can look at it right away.

If the liquid have some cruder ingredients in it, you can use an object glass with a dtch or concavity in it.

 

Looking at a fresh solid object.

A fresh solid object is usually too bulky to be looked at with illumination from below. If the object is absolutely still and small enough to fit under the objective, you can often look at it with illumination from above. If such an arrangement is not possible, you must make slices of it to be placed at a glass plate.

A practical thickness for the slice is ca 0.25 - 2.0  mm thick. The greater the magnification, the thinner must the slice be.

With some training you can make such slices with a sharp thin knife, scalpel or with an old fashion razor blade. However there are also instruments called microtomes to be bought to make such slices. These can be found from simple hand held devices to advanced scientific microtomes.

Then you lay the sliced object on the glass and lay the cover glass over it.

 

Preparing a permanent object from a liquid blending

First of all you blend some color solution in the liquid. There are colors that adhere to specific structures such as cell kernels or the cytoplasma of microorganisms. Wait for some time.

Do not use so much color that the blending get sharply colored.

You then take a drop of the colored liquid on the object glass.

The you gently warm and dry the drop.

Then lay a drop of gluing balsam on a cover glass. Such gluing balsams are to be bought. A blending of canada balsam ( a sort of natural resin ) and xylene is a commonly used glue. The blending itself is often called just Canada balsam.

Then lay the cover glass over the object with the gluing balsam towards the obejct.

You let the balsama dry, and the object is ready.

 

Preparing a permanent object from some solid material.

This is the most extensive procedure.

First you must cut a thin slice of the object.

Then lay the object on the object glass.Press it gently, but fermly to the glass. Usully the object will glue to the glass by itself.

Then set the glass with the object in a watery color solution for a couple of hours. If the object shall be colored, the color must be soluted into the water. If you use two colors, it is most practical to use two concecutive solutions, each with its own color.

Move the object over to a solution of alcohol and water, and let it stand there for a couple of hours.

Then move it over to a solution of pure alcohol and let it stand a couple of hours.

Then move it over to some liquid that is not soluble in water and let it stand a couple of hours. Usually xyelene is used, but toluene is also useful. These are toxic substances, so you must have some air ventilation.

At last lay a cover glass over the object with a drop of gluing balsam, and let the ready made object dry.


Use of colors for microscopy

There are a variety of color substances to choose from when preparing a preparate for microscopy. Two substances are however standardized for most applications, especially in medicine and pathology.

one of these is Eosine. This substance clor cytoplasmic structures and the collagen proteins in muscle fibres red.

The other is Methylene blue which colors DNA and RNA and hence the cell kernel.

The color Methyl blue which is not to be confused with methylene blue, can be used to color collagen fibres in muscles.

Fuchsine is a color frequently used to stain bacteria. Another stain frequently used for bacteria is Gentian violet. (Methyl violet, Crystal violet). It is used in the Gram's method of bactereal classification.

Bacteria that have a thick peptidoglycan layer around them are stained brightly blue-violet by gentiana violet. Peptido-glycan is a molecular network consisting of carbomydrate units chemically bound to each other. Other bacteria are only stained faintly pink. Those strongly stained are called Gram-positive and those others Gram-negative.
